Jacob Christian Fredsøe: DNA topoisomerases influence transcriptional activation of chromatin regulated genes

During the course of his PhD studies, Jacob Christian Fredsøe has investigated the role of DNA topoisomerases in transcription. By using baker’s yeast, S. cerevisiae, it has been possible for Jacob Christian Fredsøe to investigate the role of these enzymes on both a global and gene specific scale. He has demonstrated that DNA topoisomerases play a profound role during transcriptional activation of genes with a repressible/inducible mode of regulation, i.e. genes for which expression is quickly changed in response to environmental changes. An in-depth investigation of the PHO5 and GAL genes revealed that topoisomerases are required for promoter nucleosome eviction and pre-initiation complex formation, respectively during the transcription activation process.
The new research findings contribute to the understanding of how DNA topoisomerases and thereby the topological conformation of the DNA influences the transcription process.
The PhD degree was completed at the Department of Molecular Biology and Genetics, Science and Technology, Aarhus University.
