Saw Marlar: Tools for Measurement of Aquaporin Diffusion

Final adjustment of urine concentration occurs in the renal collecting duct where it is regulated by the antidiuretic hormone arginine vasopressin (AVP). Rapidly following AVP stimulation, aquaporin-2 (AQP2) is translocated from the intracellular vesicles to the apical plasma membrane instantly increasing water reabsorption and subsequent urine concentration. Water exits via basolaterally localized AQP3 and AQP4.

During her PhD studies, Saw Marlar set up an experimental system to study the short-term AVP regulation of AQP3. Using quantum dot (QD) labeling and k-space image correlation spectroscopy (kICS) approach, she observed that AQP3 is indeed regulated via changes in lateral diffusion within 15-30 minutes following AVP stimulation. The new finding indicates the occurrence of basolateral adaptations, which might aid in facilitating the AVP mediated increased water efflux. In addition, she established another experimental system to measure the lateral diffusion of AQP3 and AQP4 both in the lateral and basal membranes by use of micropatterning and kICS analysis.
